The reasoning behind the result

Three densities describe different denominators

N = σA = CfinalVfinal

N is viable cells per vessel, σ is viable cells/cm², A is measured growth area in cm², and Cfinal is viable cells per liquid-volume unit. A target of 25,000 cells/cm² in a 2 cm² vessel means 50,000 cells, regardless of final liquid volume.

A larger final liquid volume reduces cells/mL but does not reduce cells/cm² when the same cells and area are retained. When area is unknown, the tool reports the count and volume concentration without fabricating an area density.

Apply viability once, at the stock denominator

Cviable = Ctotal × viable fraction; Vstock = N / Cviable

If the instrument or count calculation already reports viable concentration, use it directly. If it reports total concentration, multiply by the separately measured viable fraction before calculating aliquots. Applying that fraction a second time would overstate the required stock volume.

The stock concentration is measured before allocation. The resulting viable-cell estimate does not predict attachment, survival after handling, recovery or growth. Those biological effects are not silently treated as a preparation allowance.

An allowance and a reserve affect opposite sides of the ledger

Vrequired = Σ(vesselsᵢ × aliquotᵢ) × (1 + allowance/100)

The allowance prepares additional material at each group's same composition; it increases both stock and final preparation volume. A reserve removes stock from what may be allocated. These are explicit planning choices, not universal laboratory recommendations.

Groups with different target densities remain separate. Their volumes can be totaled for purchasing or availability checks, but the total does not instruct you to mix all preparations together.

Some targets cannot be reached by adding medium

An aliquot larger than the desired final volume means the requested final concentration exceeds the viable stock concentration. The tool flags that row rather than reporting a negative medium volume. It does not specify a cell concentration procedure.

Even when all volumes fit, the culture conditions may be inappropriate for the material. Seeding requirements depend on the actual cell line, vessel, experiment and established protocol; this calculator never supplies a generic ideal target.

Follow the numbers

Six vessels with an entered area target

  1. At 25,000 viable cells/cm² and 2 cm² per vessel, each vessel receives 50,000 viable cells.
  2. A measured stock of 1,000,000 viable cells/mL supplies those cells in 0.05 mL. Bringing the final volume to 1 mL requires 0.95 mL cell-free medium per vessel.
  3. Six vessels require 0.30 mL stock and 5.70 mL medium. A separately entered 10% preparation allowance raises those quantities to 0.33 mL and 6.27 mL.

The 50,000 cells/vessel target stays unchanged; the allowance covers extra preparation, not a higher dose to each vessel.

How to use this calculator

  1. Enter an independently measured stock concentration. Apply viability only if that concentration counts total cells.
  2. Choose your starting target or known aliquot. Enter measured vessel areas when available; zero means area is unknown, not zero physical area.
  3. List each group, final volume and target. Enter your own extra preparation allowance and stock reserve.
  4. Inspect both the per-vessel densities and combined stock demand. A failed quantity check leaves the intended quantities visible for review.
